If you're already pushing Atlas maps, Twilight Reliquary Keys can show up when you least expect it. They're not tied to one neat little activity, which is both nice and annoying. Any endgame loot source can spit one out: monsters, chests, league-style encounters, and other bits you'd normally crack open while farming. The key matters because it opens the Reliquary Vault, a special Atlas location with a golden chest inside. That chest gives a random Foiled Unique, so players chasing unusual PoE 2 Items will usually keep an eye out for these keys while mapping.
There's no secret boss you need to farm for the standard Twilight Reliquary Key. It sits in the global endgame loot pool, so the real answer is simple: play Atlas content and clear a lot of stuff. You won't feel in control of it, because you aren't. Some players see one early. Others can run plenty of maps and get nothing. That's just how Path of Exile 2 loot tends to behave.
If you don't want to wait, use the Currency Exchange. Speak with Ange in Kingsmarch or in your Hideout, then open the exchange window. Pick "I Want" and search for Twilight Reliquary Key. After that, choose what you're paying with under "I Have." A common choice is Exalted Orbs, and the price may sit around five Exalted Orbs per key, though that number can move fast. The market's player-driven, so don't be shocked if the cost jumps during busy periods.

There's no long dungeon crawl attached to it, which makes the whole thing feel more like a gamble than a farming route.
The Vault's rewards come from Unique items selected through high-tier supporter pack submissions, so the result can vary wildly. You might hit something exciting, even a chase Unique, but most openings are more ordinary. Foiled Uniques don't gain stronger stats or hidden powers; they just have a special visual treatment on the item card. Boss Reliquary Keys work differently, since they pull from a related boss drop pool. That makes them better if you're hunting specific PoE 2 gear for sale such as a Foiled version of a boss-only Unique, but it's still a roll of the dice.
